One of the fundamental differences between these two speakers is that L100 is a 2-way design whereas the L600 is a 3-way speaker. 3-way designs utilize a midrange driver in addition to the tweeter and woofer on 2-way designs. Designing a crossover for a 3-way speaker is a complicated process and generally increases the overall cost of the speaker.

L100 features a 1" Ring Radiator Tweeter and 1 x 5.25" Polypropylene Woofer with a Crossover frequency at 2900Hz. On the other hand, the L600 features a 1" Pinnacle Ring Radiator Tweeter , a 5.25" Midrange and 2 x 7" Polypropylene Woofer with a Crossover frequency at 410Hz and 2900Hz.
Frequency Response
L100 has a frequency range of 57-38k Hz whereas L600 has a frequency range of 34-38k Hz. With a minimum frequency of 34Hz, the L600 can go significantly deeper on the low side and provide stronger bass compared to the L100's min frequency of 57Hz.

Impedance and Sensitivity
L100 and L600 have the same Impedance of 4 ohms but where they differ is the sensitivity levels. L100 sensitivity is rated at 85.5 dB and the L600's is at 86 dB.
One thing to keep in mind that the lower impedance speakers, especially the ones that are rated at 4 ohms may put more pressure on the amplifier unit comared to higher impedance speakers especially at high volume levels, so make sure the amp you pair with the L100 and the L100 can provide this impedance level.

Power Range
Power
range is the range of input power in watts RMS that a loudspeaker is designed to handle. While using an amplifier within this range ensures the nominal performance , inputting a power that is higher than the max wattage can result in a damaged speaker.
L100 can handle a max power of 300 watts RMS from the amp the whereas L100 has a max power handling value of 160 watts RMS , 140 watts less than the L600.
Keep in mind that a higher max power handling value doesn't necessarily make that a louder speaker
compared to a lower max handling speaker. Loudness or the sound level is also impacted by parameters
such as Impedance, sensitivity and system efficiencies.
Input Type and Bi-Amping / Bi-Wiring
L600 features Binding post posts which make bi-amping or bi-wiring possible. L100 has a Post which doesn't feature bi-amping/bi-wiring.
Cabinet Type and Port Position
L100 has a rear firing port whereas L600 has a down firing ports.
Physical Specs
Size of a speaker can sometimes become an important decision factor due to space constraints or in some cases purely for esthetic reasons. In this section, we are going to compare Polk L100's and Polk L600's external dimensions. Polk L100 has external dimensions of 348.0 x 201.0 x 288.0mm ( 13-11/16 x 7-7/8 x 11-5/16inch) whereas Polk L600 has external dimensions of 1182.0 x 331.0 x 426.0mm ( 46-1/2 x 13 x 16-3/4inch) .
Polk L600 is clearly the larger of the two speakers. Its body is 130mm wider, 834mm taller and 138mm deeper than Polk L100.

Base surface area of a loudspeaker may become a determining factor when the space in your room or desk is limited.
The base surface area of the Polk L100 is approximately 578.9cm2 / 89.7inch2 and base area of the Polk L600 is approximately 1410.1cm2 / 218.6inch2. The L100 requires 59% less surface area than the L600 which gives it a small advantage on placement in tight spaces.
